Callogenesis and Plant Regeneration in Peony (Paeonia × suffruticosa) Using Flower Petal Explants

چکیده

Peony is a traditional Chinese flower with significant ornamental and medicinal value. However, there are still problems, such as serious browning, difficulties in differentiation, rooting low regeneration efficiency the process of system established, which have hindered development transgenic peony technology. Establishing an efficient considered to be important goal among researchers. Here, we describe protocol for high-frequency callus induction establishment plants using petals explants. Murashige Skoog (MS) medium supplemented 2.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D), 1.5 N-(phenylmethyl)-9H-purin-6-amine (6-BA), 0.3 1-naphthylacetic (NAA) was identified best induction, achieving rate up 98.52%. The highest proliferation (234%) achieved on MS 0.2 NAA 3.0 6-BA. differentiation (34.81%) 6-BA 0.5 zeatin (ZT). 23.33% when 1/2 0.1 0.05 3-indolebutyric (IBA). After acclimation, were transferred pots, where they showed robust growth. We also observed surface structures calluses scanning electron microscopy found that characteristics hugely variable different appeared affect bud efficiency. rapid regenerating peonies petal cultures established here will create new opportunities mass reproduction genetic engineering plants.

Selection of Reference Genes for Quantitative Real-Time PCR during Flower Development in Tree Peony (Paeonia suffruticosa Andr.)

Tree peony (Paeonia suffruticosa) is a perennial plant indigenous to China known for its elegant and vibrantly colorful flowers. A few genes involved in petal pigmentation have been cloned in tree peony. However, to date, there have been few studies on the comparison and selection of stable reference genes for gene expression analysis by quantitative reverse-transcription PCR (qRT-PCR) in this ...

Chemical Profile and Antioxidant Activity of the Oil from Peony Seeds (Paeonia suffruticosa Andr.)

Peony seed oil (PSO) is a novel vegetable oil developed from the seeds of Paeonia suffruticosa Andr. The present study aimed to make an overall investigation on the chemical profile and antioxidant activities of PSO for reasonable development and utilization of this new resource food. Chemical analysis revealed that PSO was characterized by an uncommon high portion of α-linolenic acid (>38%), f...

Changes in Microbial Biomass, Activity, Functional Diversity, and Enzyme Activity in Tree Peony (Paeonia suffruticosa) Garden Soils

To understand the effects of tree peony (Paeonia suffruticosa) on soil microbiological and biochemical properties, soil samples were collected from tree peony growing sites with 3 growth years and four tree peony cultivars as well as from an adjacent wasteland in a tree peony garden at Luoyang, Henan Province of China. With the development of the tree peony garden ecosystem, soil microbial biom...

Reverse transcriptase domain sequences from tree peony (Paeonia suffruticosa) long terminal repeat retrotransposons: sequence characterization and phylogenetic analysis

Tree peony is an important horticultural plant worldwide of great ornamental and medicinal value. Long terminal repeat retrotransposons (LTR-retrotransposons) are the major components of most plant genomes and can substantially impact the genome in many ways. It is therefore crucial to understand their sequence characteristics, genetic distribution and transcriptional activity; however, no info...
